The ferry from Slussen is the most scenic option and accepts regular SL transport cards. The Tram 7 runs from city center. You can also walk across Djurgårdsbron bridge from Östermalm in about 15 minutes.

You could spend several days exploring all attractions. For a first visit, plan one full day combining 2-3 major museums (Vasa, Skansen, ABBA) with a walk or bike ride around the island. Each museum deserves 2-3 hours.
